Cis-interaction of ligands on a supported lipid bilayer affects their binding to cell adhesion receptors

چکیده

Cell adhesion on a supported lipid bilayer (SLB) functionalized with ligand proteins is widely-used cell-mimetic model for the study of trans receptor-ligand interaction that mediates adhesion, and has been shown to play an important role in unravelling molecular players cell adhesion. Experimental studies such cell-SLB systems often assume there no cis interactions between ligands. An question remains whether cis-interaction affects interaction. Using statistical-mechanical Monte Carlo simulations biologically relevant parameters, we find attractive strength 1 kBT adjacent ligands SLB can lead amplification both affinity cooperativity binding, thereby facilitating phase separation within adhering membrane. In contrast, system less sensitive repulsive SLB. Our results suggest ligand-ligand should be carefully considered experiments.

Cell adhesion to protein-micropatterned-supported lipid bilayer membranes.

A new method for constructing controlled interfaces between cells and synthetic supported lipid bilayer membranes is reported. Microcontact printing is used to define squares and grid lines of fibronectin onto glass, which subsequently direct the self-assembly of fluid lipid bilayers onto the complementary, uncoated regions of the surface. Features of fibronectin as small as 5 microm effectivel...

Supported lipid bilayer platforms to probe cell mechanobiology.

Mammalian and bacterial cells sense and exert mechanical forces through the process of mechanotransduction, which interconverts biochemical and physical signals. This is especially important in contact-dependent signaling, where ligand-receptor binding occurs at cell-cell or cell-ECM junctions. By virtue of occurring within these specialized junctions, receptors engaged in contact-dependent sig...

Bilayer edges catalyze supported lipid bilayer formation.

Supported lipid bilayers (SLB) are important for the study of membrane-based phenomena and as coatings for biosensors. Nevertheless, there is a fundamental lack of understanding of the process by which they form from vesicles in solution. We report insights into the mechanism of SLB formation by vesicle adsorption using temperature-controlled time-resolved fluorescence microscopy at low vesicle...

Functional single-cell analysis of T-cell activation by supported lipid bilayer-tethered ligands on arrays of nanowells.

Supported lipid bilayers are an important biomolecular tool for characterizing immunological synapses. Immobilized bilayers presenting tethered ligands on planar substrates have yielded both spatio-temporal and structural insights into how T cell receptors (TCRs) reorganize during the initial formation of synapses upon recognition of peptide antigens bound to major histocompatibility complex (M...

Long-Range Interaction Forces between Polymer-Supported Lipid Bilayer Membranes.

Much of the short-range forces and structures of softly supported DMPC bilayers has been described previously. However, one interesting feature of the measured force-distance profile that remained unexplained is the presence of a long-range exponentially decaying repulsive force that is not observed between rigidly supported bilayers on solid mica substrate surfaces. This observation is discuss...
